CJC-1295 is a synthetic 30-amino-acid GHRH analogue. The DAC (Drug Affinity Complex) form has a half-life of 6–8 days allowing weekly dosing; the non-DAC form has a half-life of approximately 30 minutes and is often stacked with Ipamorelin.
Overview
CJC-1295 was developed through modifications to the first 29 amino acids of endogenous GHRH: amino acid substitutions at positions 2, 8, 15, and 27 improve resistance to enzymatic cleavage by DPP-IV. The non-DAC form (also called Modified GRF 1-29 or Mod GRF 1-29) achieves a half-life of approximately 30 minutes — still much longer than native GHRH. The DAC form additionally incorporates a lysine-maleimide drug affinity complex that covalently binds to plasma albumin, extending the half-life to 6–8 days and enabling once-weekly dosing. The longer half-life comes at the cost of losing the natural pulsatile GH secretion pattern. Pairing CJC-1295 (GHRH analogue) with Ipamorelin (GHRP) is one of the most studied GH-axis stacks in research because the two compounds stimulate GH through independent pathways.
